ADAM3

Disintegrin dan metalloprotease 3, atau ADAM3, termasuk dalam keluarga protein peptidase yang disebut ADAM. Banyak dari protein ini hanya ditemukan dalam sel-sel spermatogenik, secara spesifik di bagian anterior kepala spermatozoa yang telah mengalami kapasitasi. Protein membran ini sangat penting untuk langkah-langkah krusial dalam fertilisasi, seperti migrasi sperma melalui uterus menuju oviduk serta pengikatan pada zona pelusida. Inaktivasi ADAM3 merupakan penyebab infertilitas pria.[1]

Banyak penelitian telah merinci bahwa pada tikus dengan ADAM3 null, spermatozoa gagal bermigrasi melalui persimpangan utero-tubal. Lebih lanjut, knockout gen ADAM1a yang berkerabat dekat pada tikus, yaitu reseptor estrogen yang tidak ditemukan pada spermatozoa matur, menyebabkan hilangnya ADAM3 di permukaan dan menurunnya pengikatan zona pelusida. Penelitian lain menunjukkan bahwa spermatozoa tikus ADAM3-/- gagal menunjukkan agregasi sperma-sperma, meskipun signifikansi dari hal ini masih belum diketahui.
