Ektonukleotidase
Ektonukleotidase terdiri dari keluarga enzim metabolisme nukleotida yang diekspresikan pada membran plasma dan memiliki situs aktif yang berorientasi ke luar. Enzim-enzim ini memetabolisme nukleotida menjadi nukleosida. Kontribusi ektonukleotidase dalam modulasi persinyalan purinergik bergantung pada ketersediaan dan preferensi substrat serta distribusi sel dan jaringan.[1]
Klasifikasi
Subfamili ektonukleotidase meliputi: CD39/NTPDase (ekto-nukleotida trifosfat difosfohidrolase), ekto-fosfodiesterase tipe nukleotida pirofosfatase/fosfodiesterase (NPP), alkali fosfatase, dan ekto-5’-nukleotidase/CD73.[1]
Fungsi
Ektonukleotidase menghasilkan molekul kunci untuk pemulihan purin dan pengisian kembali cadangan ATP dalam berbagai jenis sel. Derivat nukleosida yang terdefosforilasi berinteraksi dengan transporter membran untuk memungkinkan pengambilan intraseluler. Ektonukleotidase memodulasi persinyalan purinergik P2, dan reseptor P1.[2] Selain itu, ektonukleotidase menghasilkan adenosin ekstraseluler, yang meniadakan efek yang dimediasi nukleotida dan mengaktifkan reseptor adenosin, sering kali dengan efek (patofisiologis) yang berlawanan.[3]
Pembentukan adenosin
Langkah pertama dalam produksi adenosin melibatkan konversi ATP/ADP menjadi AMP. Langkah ini dilakukan oleh ENTPD1, yang juga dikenal sebagai CD39. Langkah kedua melibatkan konversi AMP menjadi adenosin. Langkah ini dilakukan oleh NT5E, yang juga dikenal sebagai CD73.[4]
