Ribonuklease (umumnya disingkat RNase) adalah jenis nuklease yang mengkatalis degradasi RNA menjadi komponen-komponen yang lebih kecil. Ribonuklease dapat dibagi menjadi endoribonuklease dan eksoribonuklease.

Degradasi RNA merupakan proses yang sangat penting pada organisme. Selain membersihkan RNA seluler yang tidak algi dibutuhkan, ribonuklease berperan penting dalam proses pematangan semua molekul RNA. Selain itu, sistem degradasi RNA aktif merupakan pertahanan pertama melawan virus-virus RNA, dan menjadi landasan strategi imun seluler yang lebih mutakhir seperti RNAi.
