Sindrom Leigh (bahasa Inggris: Leigh's disease, Leigh syndrome, Subacute Necrotizing Encephalomyelopathy, SNEM, LS) adalah kelainan neurometabolisme yang berdampak pada sistem saraf pusat. Mutasi gen pada berkas SURF1 merupakan salah satu penyebab yang paling ditemukan, dan defisiensi COX (bahasa Inggris: cytochrome c oxidase) adalah fitur yang khas ditemukan pada otot penderita SL dengan defisiensi SURF1.[1]

Saat ditemukan pertama kali pada tahun 1951, LS dijabarkan sebagai penyakit neurodegeneratif progresif dengan lesi neuropatologis pada ganglia dasar, batang otak dan kadang-kadang juga pada struktur lain dari sistem saraf pusat.

Penderita LS saat ini dirawat dengan asupan vitamin B1, atau minyak kedelai.[2]
