Universitas Prima Indonesia didirikan oleh Prof. Dr. dr. I Nyoman Ehrich Lister, M.Kes, AIFM., AIFO., Sp.KKLP (Lie Eng Kun). Cikal Bakal Universitas Prima Indonesia bermula dari Akademi Keperawatan dan Akademi Kebidanan Prima Medan yang didirikan pada tahun 2001, kemudian pada tahun 2002 berkembang menjadi Sekolah Tinggi Ilmu Kesehatan Prima Husada Medan. Pada tahun 2005, Sekolah Tinggi Ilmu Kesehatan Prima Husada Medan berkembang menjadi Universitas Prima Indonesia.
