1913年(大正2年)6月休暇帰米の際、カリフォルニアに勃発した日本移民排斥運動に遭遇、当地に留まり外国人土地法案の阻止に尽力した[6]。「外国人土地法」制定後も移民問題に従事して研鑽し、各国移民の割当制度(次節参照)を考案する[13]。翌年米国キリスト教連盟(英語版)に対日関係委員会が設置されるや幹事(同時に東洋関係委員会幹事)に就任し、「移民割当制」を提唱する著書(『The American Japanese Problem』)の刊行とともにウィルソン大統領をはじめ各界の有力者に対するロビー活動を展開してその普及につとめた[14]。1915年(大正4年)1月一旦渡日し再び帰米、同2月からは萬国キリスト教会国際親善促進同盟米国支部幹事、建設的移民法制定委員会幹事、国際正義仁愛委員会幹事などの要職に就き[14]、その後の約20年間を日本人の移民問題解決および日米親善活動に心血を注いだ[15][16]。また、国際平和会議米国代表(1914年)、パリ講和会議米国キリスト教連盟代表(1919年)、全世界キリスト教徒大会出席(1925年)などに加え、その他多くの国際問題に携って活躍し、特に極東問題に関する著述におけるその主張は識者間において権威あるものとされている[17]。
ギューリック一族は日本と非常に関わりが深く、父ルーサー、シドニー本人のほか、叔父オラメル、叔父ジョン、叔母ジュリア、次男リーズいずれもかつて日本で伝道に従事しており[5][注釈 3]、祖父母は晩年日本に移住し、共に神戸で亡くなり当地に埋葬されている[19]。また母ルイーザ(Louisa Lewis Gulick)も同じく神戸の墓地に眠っている[19]。孫のギューリック3世(英語版)については次節を参照。
